Procurement Summary

This is a new procurement. The protein structure group at the National Center for Environmental Health/Division of Laboratory Sciences/Clinical Chemistry Branch (CCB) requires an advanced instrument capable of single cell isolation, sorting, and dispensing system with integrated sample preparation solutions for mass spectrometry (MS) proteomics and transcriptomics. Both SARS-CoV-2 and Influenza A virus have continued to be significant threats to global public health. Single-cell proteomics (SCP) has provided insights into the various aspects of humoral immunity to new virus variants. SCP techniques can identify and quantify proteins expressed within individual cells for gaining a deep understanding of biological heterogeneity and contributing to therapeutic insights. It also can quantify and integrate data about cell behavior, signaling and regulatory networks with temporal information. The laboratory needs the capability to develop new methods and technology for understanding of the functional consequences of virus mutations, and deciphering the immune responses to new viruses, vaccines, and neutralizing antibodies. The laboratory will have the capacity to dissect the associations of vaccination, age, sex, disease severity, and stage with the compositional changes of immune cells in human peripheral blood mononuclear cells (PBMCs); and to identify and quantify related signal pathway, the immune dynamics of antibody responses, and correlations with disease severity or therapeutic treatment. In addition, we would be able to monitor how long the antibody and memory B-cell responses last, and whether some aberrant auto-reactive antibodies are generated etc.
