(1) Description of items being acquired: Manufactured by Berthold Technologies in NAICS CODE: 334516 – Laboratory Analytical Equipment Manufacturing • Tristar 3 for UV/VIS Abs/Lum/FL/Temp control/Cooled PMT/ICE-software, QTY 1 • BRET PACKAGE, QTY 1 • TriStar2 Additional filter slide B" (emission)", QTY 1 • cwl 495nm, fwhm 10nm, 70%, 25mm, QTY 1 • cwl 650nm, fwhm 10nm, 70%, 25mm, QTY 1 • Installation and Training, QTY 1 The National Institute of Allergy and Infectious Diseases (NIAID)’s Laboratory of Molecular Immunology (LMI) conducts basic, translational, and clinical studies related to innate and adaptive immune system function in health and disease. LMI scientists have made major contributions to our understanding of immunoregulation by chemokines and their G protein-coupled receptors, HIV pathogenesis, the NFkB family of transcription factors, mucosal immunology in the gut, reovirus and rotavirus infection in the gut, and mouse models of inflammatory bowel disease. They explore the basic properties of neutrophils, macrophages, naïve and memory T cells, and dendritic cells, as well as genetic risk factors for complex immune-mediated diseases. In LMI, current studies focus on the molecular pathogenesis of infectious and immunologic/inflammatory diseases, including West Nile virus infection, Listeria infection, Trypanosoma cruzi, Toxoplasma gondii, fungal infection, sepsis, atherosclerosis, psoriasis, inflammatory bowel disease, primary immunodeficiency disease, and cancer, working toward the goal of identifying novel therapeutic targets and strategies. The LMI is interested in procuring a Microplate Reader system that is required for performing luminometry and BRET assays in the lab. Salient Characteristics- High performance filter system for optimal sensitivity. ONE-4-ALL Optics for uncompromised performance of all detection modes (Absorbance Fluorescence Luminescence & BRET). Highly-sensitive dual mode PMT detectors in combination with the optimized ONE-4-ALL optical path design provide best-in-class luminescence detection of less than 6 amol ATP per well. Agility to move from one application to another application with a smooth transition so the system does not have to stop and restart causing information to be lost. Quick change filter technology is critical when there are 40 filters in this equipment that can be changed quickly and easily with a filter slide which allows for the equipment to run uninterrupted. The ergonomic design of the system provides full front access for all key operations. (e.g., plate loading, filter change and connecting reagents). JET injector technology for highest accuracy, speed and cell-friendliness, Broad wavelength range selection from UV through the visible range, BRET/BRET2 and NanoBRET™. Without these features another assay equipment will provide low accuracy without having reproducible results and with a higher consumption of reagents. LMI requires a highly sensitive and specialized multimode multiplate reader to perform Flash and Glow Luminescence, Absorbance, BRET, Fluorescence and FRET assays.

Waiver Rationale Summary

(3) Conclusion: Based on the above market research findings and results from previous brand name or equal synopsis and solicitations posted on SAM.gov, no domestic products that meet the requirements were identified. No domestically manufactured items that meet the Government’s need is not mined, produced, or manufactured in the United States in sufficient and reasonably available commercial quantities and of a satisfactory quality could be identified. Historically, all similar Microplate Readers were manufactured outside of the US. No agency has bought this type of equipment from a US manufacturer. The feasibility of foregoing the requirement or providing a United States-manufactured substitute has been considered and no substitutes were found. Authority is requested to acquire the above-described LB942 Tristar 3 Multimode Microplate Reader that are manufactured in Germany by Berthold Technologies for the total of $30,196.40. Also, Based on the findings detailed above, the LB942 Tristar 3 Multimode Microplate Reader are not mined, produced, or manufactured in the United States in sufficient and reasonably available commercial quantities and of a satisfactory quality. Therefore, it is hereby determined, consistent with FAR 25.103(b), that these items are nonavailable.
