TRI-COLOR MULTICHANNEL FIBER PHOTOMETRY SYSTEM

Waiver type - Procurement: Nonavailability

Agency: NATIONAL INSTITUTES OF HEALTH (7529) • Product Service Code (PSC) : 6515 | Last Modified: 08/15/2023

Procurement Summary

Tri-color multichannel fiber photometry system. This requirement aims to procure a tricolor multichannel fiber photometry system to enable researchers to take advantage of optogenetics, to explore detailed insight into the activity and behavior of neuronal populations. This technique stimulates neurons with light and measures fluorescence signals corresponding to calcium dynamics by detecting changes in the fluorescence intensity from a genetically encoded calcium indicator (GECI). Calcium dynamics related to activities and patterns in neural circuits can be measured in real-time. The technique can help develop treatments for neurological diseases and brain-related traumatic injuries. Its use of light offers many advantages over other neuromodulation methods, including a temporal resolution on the order of milliseconds, closely matching natural neuronal activity, and the stimulation of multiple neuron sets independently through different wavelengths. In fiber photometry, data is collected by analyzing the change in fluorescence (ÄF) relative to an initial baseline fluorescence (F) and observing a change in signal that corresponds to a calcium transient (ÄF/F). These indicators are typically based on fluorophores like GFP, RFP, tdTomato, mCherry, etc., of which GCaMP is the most common example. GCaMP contains Green Fluorescent Protein (GFP). As such, GCaMP exhibits the same fluorescence patterns as standalone GFP and provides key insight into calcium dynamics because GCaMP only fluoresces when bound to a calcium ion. In neurons, calcium ions regulate several important processes, including neurotransmitter release and membrane excitability. Therefore, GCaMP fluorescence is tied directly to neuronal response patterns, as depicted by the graph to the right. After searching the market, we consider the RWD tricolor multichannel fiber photometry the fit product for long-term scientific projects.

Waiver Rationale Summary

The tri-color multichannel fiber photometry system will allow researchers to detect the activity of neurons and behavior of neuronal populations in the neural circuit study. This requirement aims to procure tricolor multichannel fiber photometry system to enable researchers to take advantage of optogenetic, to explore detailed insights into the activity and behavior of neuronal populations.
